Alstonia scholaris, a prominent ethnomedicinal plant, is widely recognized for its broad-spectrum
pharmacological activities including anticancer, antimicrobial, anti-inflammatory, anti-asthmatic,
neuroprotective, and cardioprotective effects. Despite these promising properties, its clinical
application remains limited due to poor aqueous solubility, low oral bioavailability, enzymatic
degradation, and non-specific systemic distribution of its phytoconstituents such as echitamine, lupeol,
and β-amyrin. Recent advances in nanotechnology have introduced a paradigm shift in the delivery of
herbal therapeutics. Nano formulation strategies—such as polymeric nanoparticles, solid lipid
nanoparticles (SLNs), nanostructured lipid carriers (NLCs), nano emulsions, liposomes, and
micelles—significantly enhance the pharmacokinetic and pharmacodynamic profiles of A. scholaris
bioactive. These nanocarriers improve solubility, prolong systemic circulation, enable controlled and
targeted release, reduce dosing frequency, and minimize systemic toxicity. Pharmacokinetic studies
reveal marked increases in Cmax, t1/2, AUC, and relative bioavailability
of nano formulated compounds compared to their conventional counterparts. This review consolidates
recent advances in nano formulation techniques applied to A. scholaris, detailing formulation
methodologies, characterization parameters, in vitro and in vivo efficacy, and safety profiles. It also
discusses regulatory frameworks, translational barriers, and the need for standardization in herbal nano
formulations. Additionally, the integration of artificial intelligence in formulation design, scalable
green manufacturing technologies, and personalized nano-herbal therapeutics are highlighted as future
directions. In conclusion, nano formulation-based delivery offers a transformative strategy to unlock
the full therapeutic potential of Alstonia scholaris, paving the way for its translation into clinically
effective and globally accessible phytopharmaceuticals.
